It is part of normal development and helps refine neural networks. One schizophrenia hypothesis proposes that altered pruning, possibly involving immune-related mechanisms, contributes to changes in connectivity. Genetic and experimental findings have encouraged research into this possibility.

Brain development involves the formation, strengthening and reorganisation of neural connections across life. Schizophrenia research examines how developmental differences may contribute to later vulnerability. Findings usually concern group patterns and interacting processes, not a single abnormality that can diagnose an individual or determine their eventual symptoms and abilities.

The hypothesis remains more complex than the claim that schizophrenia simply involves too many connections being removed. Different regions, developmental stages and mechanisms may be involved. Measures from brain tissue or cell models do not directly reveal what occurred throughout one living person’s development. Pruning is also only one process among many. Research may identify useful treatment targets, but it does not currently provide a routine test of individual pruning or justify predicting symptoms from a simplified account. Explanations should clearly separate normal development from proposed disease mechanisms.

Synapses are sites of communication between neurons. Refinement involves changes in number, strength and organisation, so counting connections alone would not describe network function. Pruning hypotheses need evidence about location and timing as well as amount. This complexity matters when translating an experimental finding into a public explanation, especially because direct observation of these processes in living humans remains limited.

Further reading: National Institute of Mental Health (1); National Institute of Mental Health (2).

Synaptic pruning is the selective reduction and reorganisation of connections between nerve cells.
